== Construction of PAM matrices == PAM matrices were introduced by Margaret Dayhoff in 1978. The calculation of these matrices was based on 1572 observed mutations in the phylogenetic trees of 71 families of closely related proteins. The proteins to be studied were selected on the basis of having high similarity with their predecessors. The protein alignments included were required to display at least 85% identity. As a result, it is reasonable to assume that any aligned mismatches were the result of a single mutation event, rather than several at the same location. == Ecological considerations == Because of the beneficial uses of KLH, the Giant Keyhole Limpet, from which KLH is derived, has been a subject of growing concern among fisheries biologists in the state of California due to the possibility of overharvest of the species for commercial purposes. Giant Keyhole Limpets are not an abundant species to begin with, and some divers have reported a decline in their populations. In a 2006 report from the Marine Science Institute, University of California, Santa Barbara, scientists state that they are "very concerned about the emerging fishery" for Giant Keyhole Limpets and suggest the potential exists to "decimate their populations". To help assure a secure long-term source of KLH for the many Federally sponsored biomedical research programs using KLH, Federal agencies including the National Institutes of Health and the National Science Foundation have also sponsored research to establish sustainable mariculture production of the Giant Keyhole Limpet.
=== Plasma cortisol concentration === Resting plasma cortisol may be slightly elevated in affected horses, but is commonly within normal limits or below normal. Additionally, elevations may occur secondary to stress, concurrent disease, and due to individual variation. === Early history === More than two thousand years ago the Scythians lived on the banks of the river Don. Many Scythian tombs have been found in this area. Subsequently, the area was inhabited by the Khazars and the Polovtsians. From the 16th to the 18th centuries the steppes of the Don River were part of "the Wild Field" (Russian: Дикое Поле). In the late Middle Ages the area was under the general control of the Golden Horde, and numerous Tatar (especially Crimean Tatar) armed groups roamed there, attacking and enslaving merchants and settlers. The first Christians to settle on the territories around the Don were the Jassi and Kosogi tribes of the Khazar Kaghanate of the 7th to 10th centuries. After the fall of the Golden Horde in 1480, more colonists started to expand onto this land from the Novgorod Republic after the Battle of Shelon (1471), and from the neighboring Principality of Ryazan. Magnesium is a chemical element; it has symbol Mg and atomic number 12. It is a shiny gray metal having a low density, low melting point, and high chemical reactivity. Like the other alkaline earth metals (group 2 of the periodic table), it occurs naturally only in combination with other elements and almost always has an oxidation state of +2. == Structure == Epithelial tissue cells can adopt shapes of varying complexity from polyhedral to scutoidal to punakoidal. They are tightly packed and form a continuous sheet with almost no intercellular spaces. All epithelia is usually separated from underlying tissues by an extracellular fibrous basement membrane. The lining of the mouth, lung alveoli and kidney tubules are all made of epithelial tissue. The lining of the blood and lymphatic vessels are of a specialised form of epithelium called endothelium.
Shafts were drilled at the site before the test in order to accelerate sample collection after explosion, so that explosion would expel radioactive material from the epicenter through the shafts and to collecting volumes near the surface. This method was tried in two tests and instantly provided hundreds of kilograms of material, but with actinide concentration three times lower than in samples obtained after drilling. Whereas such method could have been efficient in scientific studies of short-lived isotopes, it could not improve the overall collection efficiency of the produced actinides. ==== Miniature Mass Spectrometers ==== Conventional mass spectrometers are often large as well as prohibitively expensive and complex in their operation which has led to the increased attractiveness of miniature mass spectrometers (MMS) for a variety of applications. MMS are optimized towards affordability and simple operation, often forgoing the need for experienced technicians, having a low cost of manufacture, and being small enough in size to allow for the transfer of data collection from the laboratory into the field. These advantages often come at the cost of reduced performance where MMS resolution, as well as the limits of detection and quantitation, are often barely adequate to perform specialized tasks. The integration of DMF with MMS has the potential for significant improvement of MMS systems by increasing throughput, resolution, and automation, while decreasing solvent cost, enabling lab grade analysis at a much reduced cost. In one example the use of a custom DMF system for urine drug testing enabled the creation of an instrument weighing only 25 kg with performance comparable to standard laboratory analysis.
==== Trump administration ==== On 6 February 2020, the director of the White House's Office of Science and Technology Policy requested the National Academies of Sciences, Engineering, and Medicine to convene a meeting of "experts, world class geneticists, coronavirus experts, and evolutionary biologists", to "assess what data, information and samples are needed to address the unknowns, in order to understand the evolutionary origins of COVID-19 and more effectively respond to both the outbreak and any resulting information". In April 2020, it was reported that the US intelligence community was investigating whether the virus came from an accidental leak from a Chinese lab. The hypothesis was one of several possibilities being pursued by the investigators. US Secretary of Defense Mark Esper said the results of the investigation were "inconclusive". By the end of April 2020, the Office of the Director of National Intelligence said the US intelligence community believed the coronavirus was not man-made or genetically modified. US officials criticised the "terms of reference" allowing Chinese scientists to do the first phase of preliminary research.
