A peculiarly British practice was the "loose-coupled" freight train, operated by the locomotive crew and a "guard" at the rear of the train, the successor to the brakesman of earlier times. That train type used three-link chain couplings for traction and side buffers to accept pushing forces but, since such trains were not fitted with an automatic through-train braking system, there were no pipes to connect between the vehicles. The last vehicle of the train was a heavily ballasted guard's van with its brakes controllable by a handwheel operated by the guard. The slack between vehicles coupled in this way was very convenient when starting heavy trains with a relatively low-powered locomotive on the level or on a rising gradient. On the driver's command the guard would apply his brake as hard as possible. The driver would then gently reverse to close up the wagons on to their buffers. Then the locomotive would be driven ahead, picking up the load wagon by wagon, thus giving an easy start up the gradient. Wagons of that era did not have roller bearings and the grease-lubricated plain bearings exerted considerable resistance to motion, especially on a cold day, so starting wagon-by-wagon in this way allowed the locomotive to move off with low initial resistance. The disadvantage of that convenience was that the guard could be badly thrown about as the train changed speed due to the inter-wagon gaps opening or closing. In the worst case, the jerks could break a coupling or cause a derailment.

=== Sulfide analysis === The formation of methylene blue after the reaction of hydrogen sulfide with dimethyl-p-phenylenediamine and iron(III) at pH 0.4 – 0.7 is used to determine by photometric measurements sulfide concentration in the range 0.020 to 1.50 mg/L (20 ppb to 1.5 ppm). The test is very sensitive and the blue coloration developing upon contact of the reagents with dissolved H2S is stable for 60 min. Ready-to-use kits such as the Spectroquant sulfide test facilitate routine analyses. The methylene blue sulfide test is a convenient method often used in soil microbiology to quickly detect in water the metabolic activity of sulfate reducing bacteria (SRB). In this colorimetric test, methylene blue is a product formed by the reaction and not a reagent added to the system. The addition of a strong reducing agent, such as ascorbic acid, to a sulfide-containing solution is sometimes used to prevent sulfide oxidation from atmospheric oxygen. Although it is certainly a sound precaution for the determination of sulfide with an ion selective electrode, it might however hamper the development of the blue color if the freshly formed methylene blue is also reduced, as described here above in the paragraph on redox indicator.

=== Maturation of the electrical properties of the action potential === A neuron's ability to generate and propagate an action potential changes during development. How much the membrane potential of a neuron changes as the result of a current impulse is a function of the membrane input resistance. As a cell grows, more channels are added to the membrane, causing a decrease in input resistance. A mature neuron also undergoes shorter changes in membrane potential in response to synaptic currents. Neurons from a ferret lateral geniculate nucleus have a longer time constant and larger voltage deflection at P0 than they do at P30. One consequence of the decreasing action potential duration is that the fidelity of the signal can be preserved in response to high frequency stimulation. Immature neurons are more prone to synaptic depression than potentiation after high frequency stimulation. In the early development of many organisms, the action potential is actually initially carried by calcium current rather than sodium current. The opening and closing kinetics of calcium channels during development are slower than those of the voltage-gated sodium channels that will carry the action potential in the mature neurons. The longer opening times for the calcium channels can lead to action potentials that are considerably slower than those of mature neurons. == History == Robert Guthrie is given much of the credit for pioneering the earliest screening for phenylketonuria in the late 1960s using a bacterial inhibition assay (BIA) to measure phenylalanine levels in blood samples obtained by pricking a newborn baby's heel on the second day of life on filter paper. Congenital hypothyroidism was the second disease widely added in the 1970s. Guthrie and colleagues also developed bacterial inhibition assays for the detection of maple syrup urine disease and classic galactosemia. The development of tandem mass spectrometry (MS/MS) screening in the early 1990s led to a large expansion of potentially detectable congenital metabolic diseases that can be identified by characteristic patterns of amino acids and acylcarnitines. In many regions, Guthrie's BIA has been replaced by MS/MS profiles, however the filter paper he developed is still used worldwide, and has allowed for the screening of millions of infants around the world each year. In the United States, the American College of Medical Genetics recommended a uniform panel of diseases that all infants born in every state should be screened for. They also developed an evidence-based review process for the addition of conditions in the future. The implementation of this panel across the United States meant all babies born would be screened for the same number of conditions. This recommendation is not binding for individual states, and some states may screen for disorders that are not included on this list of recommended disorders.
