Semax raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.

Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. The first four residues correspond to the ACTH(4-10) fragment, while the terminal Pro-Gly-Pro extension was added to improve metabolic stability. Its molecular formula is C37H51N9O10S and its monoisotopic mass is approximately 813.9 Da. The compound is classified as a peptide research chemical and as a registered pharmaceutical product in Russia, but it does not hold marketing approval in the European Union or the United States. Naming conventions vary across supplier catalogs.
Development of the compound took place in the Soviet Union during the 1980s, largely through peptide research groups affiliated with the Shemyakin-Ovchinnikov Institute and Moscow State University. Investigators screened fragments of adrenocorticotropic hormone for activity on the central nervous system while attempting to separate cognitive effects from hormonal ones. The shortened sequence was selected because it retained behavioral activity in animal models without stimulating corticosteroid release. Early publications described the molecule as an ACTH(4-10) analog.
Published research on this peptide originates mainly from Russian laboratories, and the wider international literature is comparatively thin. Studies have used rodent models of stroke, hypoxia, and memory tasks, with a smaller number of human trials reported. Reported outcomes include changes in attention and memory measures, along with effects on neurotrophic factor expression in some experiments. Small sample sizes, inconsistent dosing protocols, and limited independent replication are widely noted limitations, so the strength of the clinical evidence remains an open question.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.
Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.

Pharmacological accounts link semax to melanocortin signalling and to modulation of neurotrophic factor expression, particularly brain-derived neurotrophic factor and nerve growth factor. Much of this evidence comes from rodent studies using intranasal delivery, a route chosen because it allows peptides to reach the central nervous system with limited systemic exposure. Whether the same mechanisms operate in humans at comparable magnitude remains an open question. The precise receptor or receptors responsible for the reported behavioural and neuroprotective effects have not been conclusively identified.
Clinical reports describe use in ischaemic stroke, transient ischaemic attack, optic nerve conditions, and cognitive complaints, but most of these studies are small and were conducted in a single region. Systematic reviews have generally described the evidence base as limited in size and variable in methodological quality. Randomised controlled data suitable for international regulatory assessment are scarce. As a result, major treatment guidelines outside Russia do not include the peptide, and interest in it remains largely research-driven rather than routine clinical.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It is described in the literature as an analogue of the ACTH(4–10) fragment, a short N-terminal portion of adrenocorticotropic hormone that retains some neurotropic activity without the full hormonal effects of the parent peptide. The molecule carries a methionine residue at the N-terminus and two proline residues near the C-terminus, features that shape both its interactions with receptor systems and its chemical stability. The free peptide corresponds to the formula C37H51N9O10S and a molecular mass near 813.9 Da.
Development took place during the 1980s at institutes of the Russian Academy of Sciences, where peptide fragments of ACTH were screened for cognitive and neuroprotective effects. The compound received regulatory approval in Russia as a nasal preparation, marketed for conditions such as ischemic stroke, transient ischemic attacks, and optic nerve disorders. Registration in other countries has not followed. Most published clinical reports originate from a small number of Russian research groups, and independent replication outside that setting remains limited. The regulatory status therefore differs sharply between Russia and the rest of the world.
Laboratory work points toward modulation of neurotrophic signaling, particularly expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal tissue. Studies also describe effects on monoamine turnover, inflammatory mediators, and oxidative markers. These observations come mainly from animal models and cultured cells, so the causal chain in humans is not firmly established. Whether the reported molecular changes translate into measurable clinical benefit is an open question. Reviews generally present the mechanism as plausible rather than demonstrated.
By 8 May 1945, the 8th, 9th, and 15th Air Force's P-51 groups claimed some 4,950 aircraft shot down (about half of all USAAF claims in the European theater, the most claimed by any Allied fighter in air-to-air combat) and 4,131 destroyed on the ground. Losses were about 2,520 aircraft. In air combat, the top-scoring P-51 units (both of which exclusively flew Mustangs) were the 357th Fighter Group of the 8th Air Force with 565 air-to-air combat victories and the 9th Air Force's 354th Fighter Group with 664, which made it one of the top-scoring fighter groups. The top Mustang ace was the USAAF's George Preddy, whose final tally stood at 26.83 victories (a number that includes shared one half- and one third victory credits), 23 of which were scored with the P-51. Preddy was shot down and killed by friendly fire on Christmas Day 1944 during the Battle of the Bulge.

Properdin is a positive regulator of the alternative pathway through its mechanism of stabilizing the C3 convertase (C3bBb). Primary T cells, monocytes, macrophages, dendritic cells, granulocytes, and mast cells synthesize mRNA to secrete properdin. Functional properdin is a product of human liver-derived HEP G2 cells. Properdin localized in the granules of neutrophils are released by TNF, TNF/fMLP, PMA, C5a, or IL-8. Additionally, neutrophils promote complement activation upon binding of cytokines, which stabilizes the alternative pathway via release of properdin, increasing defense against microorganisms. Properdin sourced from T cells promote phagocytosis of apoptotic T cells, which is an indication of their function in recognizing and clearing out apoptotic cells. Properdin is also sourced in endothelial cells along with the other complement proteins. Endothelial gene transcripts are induced when shear stress occurs, followed by properdin release into extracellular compartments. Properdin plays an important role in tissue regulation, energy metabolism, and lipid metabolism. An experiment in properdin deficient mice concluded that properdin deficiency results in fat storage and less energy output in comparison to wild-type mice. Properdin regulates fatty acid uptake into adipose tissue.
=== Anti-aging === mTOR inhibitors may be useful for treating/preventing several age-associated conditions, including neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. After a short-term treatment with the mTOR inhibitors dactolisib and everolimus, in elderly (65 and older), treated subjects had a reduced number of infections over the course of a year. Various natural compounds, including epigallocatechin gallate (EGCG), caffeine, curcumin, berberine, quercetin, resveratrol and pterostilbene, have been reported to inhibit mTOR when applied to isolated cells in culture. As yet no high quality evidence exists that these substances inhibit mTOR signaling or extend lifespan when taken as dietary supplements by humans, despite encouraging results in animals such as fruit flies and mice. Various trials are ongoing.
The Cossack homelands were often very fertile, and during the collectivisation campaign many Cossacks shared the fate of the kulaks. According to historian Michael Kort, "During 1919 and 1920, out of a population of approximately 1.5 million Don Cossacks, the Bolshevik regime killed or deported an estimated 300,000 to 500,000". Others, such as Peter Holquist, estimate a figure of 10,000 deaths during this period, while a far greater number died during the engineered Soviet famines of 1932–33 and the Holodomor.

For example, lung resistance-related proteins, otherwise referred to as major vault proteins, are located on the nuclear membrane of cells and typically mediate nucleocytoplasmic transport. Here, the regulation of compounds between the nucleus and cytoplasm is a process that can be overexpressed by cancer cells to transport DNA-targeted chemotherapeutic drugs away from the nucleus, and essentially restrict these drugs from successfully interacting with their targets. Without reducing the total intracellular chemotherapeutic drug concentration, cancer cells have developed mechanisms to successfully dismantle drug-target interactions and confer resistance to multiple chemotherapeutic drugs. An additional mechanism of resistance interfering with drug-target interaction can be demonstrated by mutations in genes that encode for drug target proteins, such as tubulin. Broadly, tubulin proteins make up intracellular structures known as microtubules that play a fundamental role in cellular shape, division, signaling, and movement, thus making them a particular promising site of target for chemotherapeutic drugs. By altering or inhibiting microtubule dynamics in cancer cells, drugs can prohibit successfully cell division and induce the direct activation of adoptotic pathways. However, mutations that produce physical alterations in tubulin protein structure, can interfere with the several tubulin binding sites and inhibit the effective interaction and activity of these chemotherapeutic drugs.
== Methodology towards improving sequence control == The most important character of sequence-controlled polymers is its controllable sequence of polymer backbone. Nonetheless, to realize a precise sequence control and to regulate sequences in greater polymer backbones are also the most urgent issue, which needs to be addressed in the field of sequence-controlled polymers. Efforts have been made in development and optimization of methods to improve the sequence-control properties of currently existed synthetic methods and also to further brand-new methods with better synthetic efficiency and sequence control.
== Treatment == The one known curative treatment is allogeneic stem cell transplantation, but this approach involves significant risks. Other treatment options are largely supportive, and do not alter the course of the disorder (with the possible exception of ruxolitinib, as discussed below). These options may include regular folic acid, allopurinol or blood transfusions. Dexamethasone, alpha-interferon and hydroxyurea (also known as hydroxycarbamide) may play a role. Lenalidomide and thalidomide may be used in its treatment, though peripheral neuropathy is a common troublesome side-effect. Splenectomy is sometimes considered as a treatment option for patients with myelofibrosis in whom massive splenomegaly is contributing to anaemia because of hypersplenism, particularly if they have a heavy requirement for blood transfusions. However, splenectomy in the presence of massive splenomegaly is a high-risk procedure, with a mortality risk as high as 3% in some studies. In November 2011, the US Food and Drug Administration (FDA) approved ruxolitinib (Jakafi) as a treatment for intermediate or high-risk myelofibrosis. Ruxolitinib serves as an inhibitor of JAK 1 and 2. Data from two phase III studies of ruxolitinib showed that the treatment significantly reduced spleen volume, improved symptoms of myelofibrosis, and was associated with much improved overall survival rates compared to placebo. However, the beneficial effect of ruxolitinib on survival has been recently questioned.
== Overdose == Excessive doses of escitalopram usually cause relatively minor untoward effects, such as agitation and tachycardia. However, dyskinesia, hypertonia, and clonus may occur in some cases. Severe side effects of escitalopram overdose include seizures (which may be delayed), cardiovascular toxicity including QRS/QTc prolongation which can lead to arrhythmias (Torsades de pointes, ventricular fibrillation, and ventricular tachycardia), hypertension, and serotonin syndrome. Treatment of escitalopram overdoses typically involves supportive care, such as giving activated charcoal or giving benzodiazepines for seizures. Because of the risk of arrhythmias (which may be delayed) prolonged cardiac monitoring is strongly recommended.
== Post-presidency (2022–present) == At the end of his presidency, Duterte returned to Davao City and kept a low profile. Former President Gloria Macapagal-Arroyo, a political ally of Duterte, convinced him in September 2023 to participate in the Philippine political landscape. In January 2023, Duterte resumed hosting his weekly show, Gikan sa Masa, Para sa Masa (From the Masses, For The Masses), on SMNI; co-hosted by Duterte's friend and spiritual adviser Pastor Apollo Quiboloy, Duterte would speak about current political issues besetting the country. Some of Duterte's strong statements in the show, such as his opposition to the expansion of the Enhanced Defense Cooperation Agreement (EDCA) sites in the Philippines, attracted attention and were covered by the media.

=== Pleural space pressure === A pneumothorax (collapsed lung) is an abnormal collection of air in the pleural space that causes an uncoupling of the lung from the chest wall. The lung cannot expand against the air pressure inside the pleural space. An easy to understand example is a traumatic pneumothorax, where air enters the pleural space from outside the body, as occurs with puncture to the chest wall. Similarly, scuba divers ascending while holding their breath with their lungs fully inflated can cause air sacs (alveoli) to burst and leak high pressure air into the pleural space.

It is a synthetic peptide of seven amino acids. Its backbone corresponds to a fragment of adrenocorticotropic hormone extended at the carboxyl end. The material is distributed as a lyophilized solid for laboratory use.
The extension was introduced to slow enzymatic breakdown of the peptide in biological systems. Natural ACTH fragments are degraded rapidly by peptidases. Adding the tripeptide was intended to extend the window during which the molecule remains intact.
Most published work consists of animal experiments conducted in a small number of laboratories. Human data are limited in number and size. Whether the observed effects translate into reliable clinical benefit is still unresolved.
The sequence is Met-Glu-His-Phe-Pro-Gly-Pro, a seven-residue peptide. The first four residues correspond to the ACTH(4-10) fragment, and Pro-Gly-Pro is added at the C-terminus. N-acetylated versions also appear in the literature and differ in mass.
