==== Myelopathy ==== Copper deficiency myelopathy in humans was discovered and first described by Schleper and Stuerenburg in 2001. They described a patient with a history of gastrectomy and partial colonic resection who presented with severe tetraparesis and painful paraesthesias and who was found on imaging to have dorsomedial cervical cord T2 hyperintensity. Upon further analysis, it was found that the patient had decreased levels of serum coeruloplasmin, serum copper, and CSF copper. The patient was treated with parenteral copper and the patient's paraesthesias did resolve. Since this discovery, awareness of copper-deficiency myelopathy and its treatment has increased. Patients typically present difficulty walking (gait difficulty) caused by sensory ataxia (irregular muscle coordination) due to dorsal column dysfunction or degeneration of the spinal cord (myelopathy). Patients with ataxic gait have problems balancing and display an unstable wide walk. They often feel tremors in their torso, causing sideways jerks and lunges. In brain MRI, there is often an increased T2 signalling at the posterior columns of the spinal cord in patients with myelopathy caused by copper deficiency. T2 signalling is often an indicator of neurodegeneration. There are some changes in the spinal cord MRI involving the thoracic cord, the cervical cord, or sometimes both. Copper deficiency myelopathy is often compared to subacute combined degeneration (SCD).
Later, the computers identified four additional missiles in the air, all directed towards the Soviet Union. Chasing the Scream: The First and Last Days of the War on Drugs was published in the UK on January 15th, 2015, followed by the US on January 20th the same month. Chasing the Scream critiques the global prohibition of illicit narcotics. Hari argued that most addictions are functional responses to experiences and a lack of healthy supportive relationships, rather than a simple biological need for a particular substance. Due to the previous scandals, Hari put the audio of some interviews conducted for Chasing the Scream online. Writer Jeremy Duns criticised instances where quotes were inaccurately transcribed or misrepresented, stating that out of a sample of dozens of clips, "in almost all cases, words in quotes had been changed or omitted without being noted, often for no apparent purpose, but in several cases to subtly change the narrative." In a review for New Matilda, Michael Brull expressed reservations about Hari's citational practices and highlighted contradictions between the narrative in Chasing the Scream and a 2009 article by Hari. ==== Vabicaserin and aripiprazole ==== Vabicaserin has a high affinity for 5-HT2C receptors and low affinity for 5-HT2B and 5-HT2A receptors. Vabicaserin is a full agonist with approximately 4-fold greater selectivity for 5-HT2C over these related receptors, in terms of binding affinity. Vabicacserin is a full agonist in stimulating the 5-HT2C receptor; it was discovered when a class of tetrahydroquinoline-fused diazepines were being researched as possible potent 5-HT2C receptor agonists. As of 2012, vabicaserin is in clinical trials for the treatment of schizophrenia. Long-term administration of vabicaserin significantly decreased the number of spontaneously active mesocorticolimbic dopamine neurons without affecting nigrostriatal dopamine neurons, consistent with effects of atypical antipsychotic agents. The outcome of clinical studies for vabicaserin may reveal whether 5-HT2C receptors can be possible targets for the treatment of schizophrenia.
