=== Canonical ensemble (NVT) === In the canonical ensemble, amount of substance (N), volume (V) and temperature (T) are conserved. It is also sometimes called constant temperature molecular dynamics (CTMD). In NVT, the energy of endothermic and exothermic processes is exchanged with a thermostat. A variety of thermostat algorithms are available to add and remove energy from the boundaries of an MD simulation in a more or less realistic way, approximating the canonical ensemble. Popular methods to control temperature include velocity rescaling, the Nosé–Hoover thermostat, Nosé–Hoover chains, the Berendsen thermostat, the Andersen thermostat and Langevin dynamics. The Berendsen thermostat might introduce the flying ice cube effect, which leads to unphysical translations and rotations of the simulated system. Ferroptosis has also been demonstrated in canine cancer cell models. There have been limited studies in other model organisms such as D. melanogaster. Elements related to components of the ferroptosis pathway have been identified in archaea, bacteria, and fungi, though it is unclear the extent to which ferroptosis occurs in these organisms. Further studies in this area may reveal an ancient origin for ferroptosis. Unlike other forms of cell death, ferroptosis has been shown to propagate between cells in a wave-like manner. This phenomenon is promoted by secretion of galectin-13 during ferroptosis. Mechanistically, galectin-13 binds to CD44, inhibiting CD44-mediated membrane localization of SLC7A11.
== Medical uses == Tiratricol is indicated for the treatment of peripheral thyrotoxicosis in people with monocarboxylate transporter 8 (MCT8) deficiency (Allan-Herndon-Dudley syndrome), from birth. Thyrotoxicosis in MCT8 deficiency is caused by a mutation (change) in the gene that provides instructions for making a protein called MCT8. This protein normally transports the thyroid hormone T3 into cells of various tissues and organs, including those of the brain. Due to the gene mutation, MCT8 does not work properly and so T3 cannot enter the brain cells, causing problems with brain and muscle development. It also leads to a buildup of T3 in other parts of the body, causing problems due to too much thyroid hormone (hyperthyroidism). Tiratricol, is very similar to T3 but it does not need MCT8 to move in and out of cells. Tiratricol is therefore expected to enter the body’s cells in people with MCT8 deficiency and replace the missing T3 hormone. This helps restore normal thyroid hormone activity in the body and reduce symptoms of peripheral thyrotoxicosis. MCT8 deficiency is a rare genetic disorder that primarily affects males. The condition is caused by a faulty gene that provides instructions for making the MCT8 transporter, a critical protein responsible for carrying thyroid hormone into the brain. Because thyroid hormone cannot cross the blood-brain barrier without the MCT8 transporter, the brain receives too little of this hormone, while excessive levels of the hormone build up in the bloodstream.

Uterine prolapse occurs when there is a disruption to any of the structures mentioned above that help hold the uterus in place. Weakening of the levator ani muscles can occur during vaginal childbirth, in which portions of the muscle can detach from the bony pelvis, or through age-related changes to musculature, and this can lead to a loss of support for the uterus. Pregnancy, vaginal childbirth, or injury can also stretch and weaken the uterosacral ligaments, leading to poor suspension or positioning of the uterus so that it is no longer supported by pelvic floor muscles. Problems with the vaginal wall, such as trauma or loss of smooth muscle support in the wall, can lead to the uterus collapsing downward due to a loss of support. When the uterus prolapses, it also drags the upper portion of the vagina (the apical vagina) along with it due to its anatomic relationship with the apical vagina. Additionally, the pelvic musculature and connective tissues are estrogen sensitive and respond to changes in estrogen level. Estrogen deficiency, which can occur during menopause, can affect the production of collagen that is needed to build connective tissue that makes up ligaments and fascia, which can contribute to uterine prolapse. This is also a reason that connective tissue disorders can predispose certain people to uterine prolapse.
== Biography == Born at Port Stanley in 1942 to New Zealanders Harold and Moya (née Boak) Baker, he spent his early life in the Falkland Islands, where his father was the superintendent of education. The family returned to New Zealand in 1948. He was educated at King's College, Auckland from 1956 to 1960. After studying chemistry at the University of Auckland, completing his PhD in 1967, he conducted postdoctoral research on the structure of insulin with Nobel laureate Dorothy Hodgkin at the University of Oxford. He then took up an academic post at Massey University, where he determined the structure of the kiwifruit enzyme actinidin. In 1997 he moved back to the University of Auckland where he became professor of structural biology and later direct of the Maurice Wilkins Center for Molecular Diversity. He also served as president of the International Union of Crystallography between 1996 and 1999. Baker was elected a Fellow of the Royal Society of New Zealand in 1987, and won the society's Hector Medal in 1997. He was awarded the Rutherford Medal, the highest honour in New Zealand science, in 2006. In the 2007 Queen's Birthday Honours, he was appointed a Companion of the New Zealand Order of Merit, for services to science.
=== Limitations of non-invasive electrical and magnetic methods === Brain tissue stimulation using non-invasive electrical and magnetic methods raises several concerns, including the following: The first issue is the uncertain dose for healthy stimulation. While neurophysiology lacks knowledge about the nature of such a treatment of nervous diseases at the cellular level, non-invasive electrical and magnetic therapies involve excessive exposure of the brain to an intense field, which is several times and even orders of magnitude higher than natural electromagnetic fields in the brain. Another significant challenge of non-invasive electrical and magnetic methods is to localize the effect of stimulation on specific neuronal networks that need to be treated. We still need to gain knowledge about mental processes at the cellular level. Neuronal correlates of cognitive functions are still intriguing questions for contemporary research. Non-invasive electrical and magnetic brain tissue stimulation targets a large area of poorly characterized tissue. Therefore, it is unclear whether electrical and magnetic fields reach only the neuronal networks of the brain that need treatment. Again, these methods involve excessive exposure to intense electrical and magnetic fields several times and even orders of magnitude higher than natural ones in the brain. However, non-invasive electrical and magnetic brain tissue methods cannot target only the neuronal networks that need to be treated. == Disease relevance == In humans, there has been a significant increase in disease from pathogenic fungi, so developing anti-fungal drugs is an important biochemical task. As homoserine dehydrogenase is found mainly in plants, bacteria, and yeast, but not mammals, it is a strong target for antifungal drug development. Recently, 5-hydroxy-4-oxonorvaline (HON) was discovered to target and inhibit HSD activity irreversibly. HON is structurally similar to aspartate semialdehyde, so it is postulated that it serves as a competitive inhibitor for HSD. Likewise, (S) 2-amino-4-oxo-5-hydroxypentanoic acid (RI-331), another amino acid analog, has also been shown to inhibit HSD. Both of these compounds are effective against Cryptococcus neoformans and Cladosporium fulvum, among others. In addition to amino acid analogs, several phenolic compounds have been shown to inhibit HSD activity. Like HON and RI-331, these molecules are competitive inhibitors that bind to the enzyme active site. Specifically, the phenolic hydroxyl group interacts with the amino acid binding site.

== Regulation of expression and activity == The formation of guanidinoacetate is normally the rate-limiting step of creatine biosynthesis. Consequently, the AGAT reaction is the most likely control step in the pathway, a hypothesis that is supported by a great deal of experimental work. Most important in this respect is the feedback repression of AGAT by creatine, the end-product of the pathway. Cyclocreatine, N-acetimidoylsarcosine, and N-ethylguanidinoacetate display repressor activity like creatine as well. L-Arginine and guanidinoacetate have only "apparent" repressor activity. They exert no effect on AGAT expression by themselves but are readily converted to creatine, which then acts as the true repressor. It has been suggested that AGAT activity in tissues is regulated in a number of ways including induction by growth hormone and thyroxine, inhibition of the enzyme by ornithine, and repression of its synthesis by creatine. Sex hormones may regulate the activity of AGAT. Treatment of male rats with testosterone propionate increases AGAT activity. In contrast, estrogen treatment decreases AGAT activity and induces weight loss. It is currently unclear whether the changes in the level of AGAT transcript results from altered mRNA stability or enhanced transcriptional rate. Their results showed not only an intense uniform fluorescence along the fiber axis but also an intense uniform composite fluorescence when the fiber was decorated with two different fluorophores in a 1:1 ratio. To prove the functional azide group could be decorated with a clinically relevant molecule, the researchers attempted to decorate the fiber with glycidyl propargyl ether (an acid-labile linker) and bound Levofloxacin (a gram-positive targeting antibiotic) to it using an ester bond between the epoxide carboxylate groups respectively. They conducted an inhibition zone assay with the functionalized silk fibers against E. Coli NCTC 12242 bacteria where each factor level contained LB media. Their results showed a successful functionalization of the Levofloxacin decorated fiber which maintained an antibiotic persistence across a 3.5 cm radius for 120 hours and a cell density ~50% of other factor levels (LB media only, unfunctionalized silk, and Levofloxacin doped silk) with p ≤ 0.01. A maximum sustained release of Levofloxacin from the fiber of 5 days was achieved.
It found that pseudoephedrine had a small risk of insomnia and this was the only side effect that occurred at rates significantly different from placebo. Insomnia occurred at a rate of 5% and had an odds ratio (OR) of 6.18. Other side effects, including headache and hypertension, occurred at rates of less than 4% and were not different from placebo. Tachyphylaxis is known to develop with prolonged use of pseudoephedrine, especially when it is re-administered at short intervals. There is a case report of temporary depressive symptoms upon discontinuation and withdrawal from pseudoephedrine. The withdrawal symptoms included worsened mood and sadness, profoundly decreased energy, a worsened view of oneself, decreased concentration, psychomotor retardation, increased appetite, and increased need for sleep. Pseudoephedrine has psychostimulant effects at high doses and is a positive reinforcer with amphetamine-like effects in animals including rats and monkeys. However, it is substantially less potent than methamphetamine or cocaine.
